Genome-wide association studies

چکیده
Genome-wide association studies (GWAS) test hundreds of thousands genetic variants across many genomes to find those statistically associated with a specific trait or disease. This methodology has generated myriad robust associations for range traits and diseases, the number is expected grow steadily as GWAS sample sizes increase. results have applications, such gaining insight into phenotype’s underlying biology, estimating its heritability, calculating correlations, making clinical risk predictions, informing drug development programmes inferring potential causal relationships between factors health outcomes. In this Primer, we provide reader an introduction GWAS, explaining their statistical basis how they are conducted, describe state-of-the art approaches discuss limitations challenges, concluding overview current future applications results. Uffelmann et al. key considerations best practices conducting genome-wide (GWAS), techniques deriving functional inferences from in understanding disease architecture. The Primer also provides information on data sharing discusses important ethical when considering populations data.

Genome-wide Association Studies
Progress in probabilistic generative models has accelerated, developing richer models with neural architectures, implicit densities, and with scalable algorithms for their Bayesian inference. However, there has been limited progress in models that capture causal relationships, for example, how individual genetic factors cause major human diseases. In this work, we focus on two challenges in par...

Genome-wide association (GWA) studies are best understood as an extension of candidate gene association studies, scaled up to cover hundreds of thousands of markers across the genome in samples usually of several thousand cases and controls. The GWA approach allows the detection of much smaller effect sizes than with previous linkage-based genome-wide studies. However, this sensitivity makes th...

A host of data on genetic variation from the Human Genome and International HapMap projects, and advances in high-throughput genotyping technologies, have made genome-wide association (GWA) studies technically feasible. GWA studies help in the discovery and quantification of the genetic components of disease risks, many of which have not been unveiled before, and have opened a new avenue to und...

Genome-wide association (GWA) studies are a powerful tool for understanding the genetic underpinnings of human disease. In this article, we briefly review the role and findings of GWA studies in type 1 diabetes, stature, pubertal timing, obesity, and vitamin D deficiency. We then discuss the present and future implications of these findings with regards to disease prediction, uncovering basic b...
